Combined inhibition of p97 and the proteasome dramatically affects ER configuration. ( A ) RPMI 8226 myeloma cells were stained with ER Tracker Blue-White DPX following treatment for 24h with bortezomib (BTZ; 5nM), Eer1 (5µM), or both. Representative confocal microscopic images show minor ER alterations after BTZ treatment, transformation of tubule-lamellar into globular ER structures after treatment with Eer1, and widespread ER vacuolisation after dual treatment. ( B ) Representative electron microscopic images of OPM-2 cells after treatment with BTZ (5nM), Eer1 (5µM), or BTZ and Eer1, for 24h. Arrows indicate classical ER in control cells, black arrowheads indicate moderately dilated and disrupted ER in Eer1-treated cells, and open arrowheads indicate vacuolised ER with reduced ribosomes on the cytosolic ER surface. Another cell treated with Eer1 and BTZ is shown at lower magnification (right panel). Areas of dilated perinuclear space are indicated by asterisks. Nu, nucleus.

Multiplex immunofluorescence staining in sequential kidney biopsies. Using two multiplex immunofluorescence assays CD38-CD138-CD27 (A, C, E, G) and CD38-CD19-CD16 (B, D, F, H) revealed changing phenotypes of immune cells across consecutive biopsies. Representative images, including magnified regions of interest with separate detection channels were shown for 2nd (A, B), 3rd (C, D) and 4th (E, F) biopsies using a Zeiss LSM710 confocal laser scanning microscope (Zeiss, Oberkochen, Germany) with a 20x objective. From every section, 4-5 representative images per biopsy were subsequently analyzed for the frequency of immune cell subsets using QuPath software showing the mean cell counts per mm 2. White arrows indicate CD16+ cells (B). *in addition, CD38+ material colocalized with diffuse DAPI (4',6-diamidino-2-phenylindole) staining, indicating broken or lysed CD38+ cells.

SEC24A&B-depleted secretory MM cells accumulate misfolded proteins and increase the size of the ER. a, b TPE-MI fluorescence was measured by flow cytometry after staining cells with 50 uM TPE-MI for 30 min. The data show the percentage change in median fluorescence intensity compared to the siRNA control condition for a AMO-1 and b NCI-H929 cells. Means +/-SEM are shown (n = 3-5 measurements). Statistical significance was measured with one-way ANOVA followed by Dunnett's test (*** P <0.001; * P < 0.05). Representative histograms are shown on the right side of each graph with the same color code. c After 32 h of siRNA-mediated transfection of MM cell lines, cells were treated with vehicle (DMSO), 2 uM KIRA6, or 40 uM STF083010 for an additional 16 h.
